I disturbi psichiatrici sono malattie altamente invalidanti e con elevatissimo impatto sociale, la cui eziologia è dovuta ad una combinazione di anomalie genetiche di suscettibilità e fattori ambientali.Il progetto si propone di determinare le basi genetiche dei disturbi dell'umore e d'ansia relativamente ai meccanismi fisiopatologici, risposta ai trattamenti farmacologici e decorso. Le difficoltà presentate da questo tipo di studi saranno affrontate con un approccio multidisciplinare volto ad integrare l'aspetto clinico, neurofisiologico, farmacologico con quello genetico. Le ricadute in campo clinico consisteranno in trattamenti più mirati ed, in ultima analisi, in trattamenti innovativi.

Obiettivi del presente progetto sono la raccolta dei dati clinici, l'analisi genetica, la centralizzazione dei dati, l'analisi dei dati e la pubblicazione.In dettaglio gli obiettivi finali sono:
1) Studio della risposta farmacologica (SSRI). Il presente studio si prefigge di indagare i determinanti genetici della risposta farmacologica e di poter predire, in base all'assetto genetico e ad aspetti clinici, il profilo di efficacia dei trattamenti.
2) Studio con marker neuropsicologici e CO2. Il progetto si prefigge di studiare le caratteristiche genetiche di sottofenotipi dei disturbi dell'umore e d'ansia identificati sulla base di caratteristiche neurofisiologiche e neuropsicologiche, e la loro correlazione con aspetti clinico-terapeutici.
3) Studio della sintomatologia e dell'andamento di malattia. Studio della componente genetica in relazione alla sintomatologia dei soggetti affetti ed al diverso andamento temporale (time-course).
4) Studio di imaging molecolare in vivo. Reclutamento dei soggetti per la caratterizzazione dell'espressione e del coinvolgimento dei recettori serotoninergici corticali nella depressione maggiore e loro correlati clinici, genetici e farmacologici.
