The present project focuses its attention on the mechanisms by which the interactions among molecules able to recognise themselves in a specific way influence cellular functionality. The project will be carried out not only by studying the cellular effects of the interaction between wild type molecules, but also by investigating the consequences of the inhibition of these specific interactions and the consequences of the modifications obtained by mutagenesis of the involved proteins and their molecular partners. The project is therefore focused on establishing the exact correlation between the structure of interacting molecules, the type of the interaction and the consequence of such interaction on cellular functionality.The project is divided in four workpackages, each of them regarding specific topics in cellular physiology.
The first WP will use as an experimental model the study of the interactions between growth factors (GF) and their receptors. It will be focused on the possibility to modify specific domain of GF by genetic engineering in order to obtain useful tools for therapeutic purposes. Moreover, the project will investigate the role of semaphorine, a protein involved in angiogenesis that is synthesised by GF-activated endothelial cells.
The second WP will focus its attention on the adhesive receptors binding to soluble or cell surface bound ligands. The attention will be placed on the mutual influences between the adhesion mediated by>>>

The general aim of this project is to point out different experimental models to study the cellular functions as a consequence of a specific molecular recognition. Sometimes the interactions involve only two components, while, more frequently, they require several molecular structures to achieve the correct cellular function.The proponent group therefore non only will investigate cellular events consequent to the interaction between two specific molecular components; it will also consider the structural characteristics of the interactions and their correlations with cell function consequences. Moreover it will be investigated the mechanisms by which the primary interaction and the cellular function are modified and regulated by the binding of other molecular components.
These projects are supported by the deep experience of the proponent groups in different fields of biological research that fit with the general topic of the present proposal. This experience results also in the availability in the different research units of advanced experimental model like mutant cell lines, transgenic mice and monoclonal antibodies.
On the basis of the above considerations the project considers four fields of biological research, very important for basic science but also strongly correlated with the pathogenesis of different diseases and with the development of diagnostic and therapeutic possibilities.
