L'ATP rilasciato nel milieu extracellulare in seguito a processi quali l'attivazione endoteliale o l'aggregazione piastrinica, è in grado di agire su recettori specifici transmembrana denominati recettori P2, regolando numerose funzioni cellulari: chemiotassi, rilascio di citochine o di costituenti lisosomiali, generazione di radicali dell'ossigeno e di ossido nitrico, regolazione del trasporto di glucosio. I recettori P2 sono a loro volta suddivisi in due sottotipi (P2X e P2Y) in base alle caratteristiche molecolari; tra i recettori P2X, il P2X7 appare svolgere un ruolo particolarmente importante nel modulare il rilascio di citochine, la proliferazione e morte cellulare, l'attivazione di fattori di trascrizione nucleare. Recentemente è' stato dimostrato che P2X7 è espresso dall'epitelio di carcinoma prostatico e da linee cellulari di carcinoma colorettale, nonché in alcuni tumori cutanei; inoltre è stato segnalato che linfociti di pazienti portatori di forme evolutive di leucemia linfatica cronica a cellule B sono caratterizzate da un aumento dell'espressione e dell'attività funzionale di P2X7.In un nostro precedente progetto PRIN cofinanziato nel 2002 abbiamo per la prima volta caratterizzato il sistema purinergico nei tireociti umani primari, evidenziando il suo ruolo nella regolazione della sintesi e del rilascio di IL-6. Durante quegli esperimenti, abbiamo casualmente osservato una marcata iperespressione di P2X7 in alcuni campioni istologici di carcinoma >>>
